MicroRNAs 与胃癌早期诊断、化疗耐药及预后的作用机制研究进展

摘要:

胃癌是世界范围内最常见的恶性肿瘤之一。由于胃癌早期缺乏特异性临床症状,患者确诊时常已处于进展阶段。进展 期胃癌往往需要联合化疗,但由于化疗多药耐药的存在,常导致疗效不佳。而确诊晚、化疗耐药等一系列问题严重影响了胃 癌的预后。因此,提高胃癌的早期诊断率,监测预后情况,清晰了解胃癌耐药的相关机制并进行靶向治疗对提高胃癌整体生 存率至关重要。大量研究表明,microRNAs(miRNAs)参与胃癌的侵袭、转移、预后及化疗耐药等。此篇综述总结了近期发现的 与胃癌诊断、预后、耐药等相关的miRNAs,如miRNA-627、miRNA-629、miRNA-652(诊断),miRNA-10b、miRNA-21、miRNA-223、miRNA-338、let-7a、miRNA-30a-5p、miRNA-126(预后),miRNA-181b 和miRNA-497(耐药),有利于胃癌相关生物学标志物研究的发展。

    The research progress of the mechanism of microRNAs in early diagnosis, chemotherapy resistance

    and prognosis of gastric cancer

    ABSTRACT: Gastric cancer is one of the most common malignant tumors worldwide. Since the gastric cancer is lack of specific symptoms in the early stage, patients are generally diagnosed at a rather advanced stage. Patients with advanced gastric cancer are subject to receiving surgery combining with chemotherapy. However, the acquisition of drug resistance always leads to poor curative effects. All of these seriously influence the prognosis of gastric cancer. Thus, it is significantly important to enhance the early diagnosis, screening and understanding of mechanisms of drug resistance, as well as comducting targeted treatment for enhancing the overall survival. Numerous researches indicates that microRNAs (miRNAs) play a pivotal role in the invasion, metastasis, chemoresistance and prognosis of gastric cancer. This review summarizes some recently identified miRNAs, which are related with the diagnosis, chemotherapy and prognosis of gastric cancer such as miRNA-627, miRNA-629, miRNA-652 (diagnosis), miRNA-10b, miRNA-21, miRNA-223, miRNA-338, let-7a, miRNA-30a-5p and miRNA-126 (prognosis), miRNA-181b, miRNA-497 (chemotherapy). This will be beneficial to the study development of biomarkers for gastric cancer.
