大剂量糖皮质激素对骨髓间充质干细胞成骨分化的 MiroRNA的调控作用

摘要:
目的 探究不同浓度糖皮质激素对骨髓间充质干细胞(BMSC)成骨分化的MicroRNA 的调控作用。方法 利用10-8 mmol/L 及10-10 mmol/L 两组浓度地塞米松对BMSC 进行诱导,对比研究诱导7、14 d 时骨钙蛋白、骨桥蛋白及碱性磷酸酶的浓度变 化。结果 诱导7 d 时,两组的骨桥蛋白浓度均呈现升高趋势,且10-10 组骨桥蛋白浓度明显高于10-8 组(P<0.05)。诱导14 d 时,
10-8 组骨钙蛋白及骨桥蛋白的浓度降低,而10-10 组骨钙蛋白及骨桥蛋白的浓度升高,且10-10 组骨钙蛋白的浓度及骨桥蛋白 明显高于10-8 组(P<0.05)。诱导14 d 时,10-8 组碱性磷酸酶浓度升高,10-10 组碱性磷酸酶浓度降低,10-10 组明显低于10-8 组(P<0.05)。结论 高浓度的糖皮质激素诱导MicroRNA 调控BMSC 向脂肪细胞分化,抑制其向骨细胞分化。

Regulatory role of high doses of glucocorticoids on MiroRNA in osteogenic differentiation of bone mesenchymal stem cells

ABSTRACT: Objective To explore the regulation of different doses of glucocorticoids on MiroRNA in osteogenic differentiation of bone mesenchymal stem cells. Methods BMSC were induced by 10 -8 mmol/L and 10 -10 mmol/L of dexamethasone in two groups, and the concentration changes of osteocalcin, osteopontin and alkaline phosphatase at induced 7 and 14 days were compared. Results At induced 7 days, the concentrations of osteopontin in the two groups increased, and that in the 10-10 group was higher than the 10-8 group (P<0.05). At induced 14 days, the concentrations of osteocalcin and osteopontin in the 10-8 group decreased, and those in the 10-10 group increased, and those of the 10-10 group higher than the 10-8 group (P<0.05). At induced 14 days, the concentration of alkaline phosphatase in the 10-8 group increased, that in the 10-10 group decreased, and that in the 10-10 group was lower than the 10-8 group (P<0.05). Conclusion High concentration of glucocorticoids induced MicroRNA to regulate the differentiation of BMSC into adipocytes, and inhibit the differentiation of osteoblasts into adipocytes.
